胡黄连对脑缺血大鼠神经生长因子表达的影响

摘要 目的观察缺血再灌注(I/R)模型大鼠脑内神经生长因子(NGF)的变化,探讨胡黄连对神经元的保护作用。方法参照线栓法制作大鼠脑I/R模型,胡黄连干预,免疫组化方法观察皮层、纹状体NGF表达。结果模型组缺血侧皮层区、纹状体区于I/R3d时NGF表达至高峰,7d后逐渐下降,但各时间点与假手术组比较,均有显著差异(P<0.01);胡黄连治疗组大鼠脑皮层与纹状体NGF明显高于正常组和模型组(P<0.01)。结论胡黄连对脑I/R损伤神经元有保护作用,其机制可能与上调脑内NGF表达有关。 Objective To observe the expression of nerve growth factor (NGF) in ischemic reperfusion (I/R) rat brain and investigate its mechanism. Methods The cerebral I/R models in rats were made by intraluminal thread method and interfered by picrorhizae. The expression of NGF in cortex and striatum were determined by immunohistoehemistry. Results In model group, the expression of NGF peaked at 3 day on ischemic cortex and striatum and descended gradually after 7 days. But compared to model group, there was significant difference at each time point ( P 〈 0. 01 ). In rat cortex and striatum of picrorhizae treated group, the expression of NGF positive neurons was higher than that of sham operated group and model group significantly ( P 〈0.01 ). Conclusions Picrorhizae can protect the neuron from I/R injury by increasing the expression of NGF in brain.
