依折麦布对兔动脉粥样硬化作用的实验研究 被引量:6

摘要 目的探讨依折麦布对新西兰兔动脉粥样硬化斑块的影响。方法将24只雄性新西兰白兔随机分为对照组、模型组、依折麦布干预组,每组8只。对照组喂普通饲料,模型组喂高胆固醇饲料(高胆固醇饲料由普通饲料加2%胆固醇配制),依折麦布干预组在高胆固醇饲料中加入5mg.kg-1.d-1依折麦布,连续饲喂12w,以镜下见到内膜增生和明显的脂斑突起作为模型制备成功的标志。HE染色观察主动脉病理形态学改变,油红O染色和高效液相色谱观察主动脉平滑肌细胞内脂质蓄积情况,RT-PCR检测主动脉平滑肌细胞脂质代谢相关基因CD36、周脂素和三磷酸腺苷结合盒转运子A1(ATP-binding cassette transporter A1,ABCA1)mRNA的变化;western-blot观察细胞膜ABCA1、蛋白激酶Cα(protein kinase Cα,PKCα)蛋白表达;PepTag○RAssay法检测细胞膜PKC活性。结果依折麦布可使新西兰兔动脉粥样硬化病变区内膜变薄、脂质蓄积减少进而逆转斑块的演进;相对于50μg/ml oxLDL处理24h的新西兰兔主动脉平滑肌细胞,3μmol/L依折麦布可有效逆转oxLDL诱导的平滑肌源性泡沫细胞的形成,使细胞内CE/TC比值降至39.6%。依折麦布可显著下调平滑肌细胞CD36、perilipin和PKCα的表达,上调胞膜ABCA1的表达。3μmol/L依折麦布可下调主动脉平滑肌细胞胞膜PKC活性至基线附近。结论依折麦布可能通过对PKC/CD36/perilipin/ABCA1这些脂质蓄积调控子的影响,发挥抗动脉粥样硬化效应。
