摘要 目的探讨两种脂肪因子——脂联素和瘦素对急性肝损伤小鼠肝细胞凋亡的干预作用。方法雄性BALB/c小鼠26只随机分为4组:刀豆蛋白A(ConA)组8只:静脉注射ConA;脂联素组5只:在静脉注射ConA前腹腔内注射脂联素;瘦素组5只:在静脉注射ConA前腹腔内注射瘦素;对照组8只:静脉注射生理盐水。注射ConA8h后处死小鼠,HE染色观察肝组织的炎症程度,TUNEL法和流式细胞术检测肝细胞凋亡率。结果与ConA组比较,脂联素组肝组织炎症程度明显减轻,血清ALT水平和肝细胞凋亡率均降低(P<0.01),瘦素组肝细胞凋亡率升高(P<0.01),而肝组织炎症变化及血清ALT水平两组无差异。结论脂联素可抑制肝细胞凋亡,减轻急性肝损伤;而瘦素则促进肝细胞凋亡,对炎症无明显影响。 Objective To investigate the interference role that two kinds of adipokines,adiponectin and leptin, exert to hepatocellular apoptosis in mice with acute liver injury induced by Concanavalin A (ConA). Methods Twenty-six male BALB/c mice were divided into four groups randomly: ConA group (8 mice), adiponectin group (5 mice), leptin group (5 mice) and control group (8 mice). Mice were sacrificed 8 hours after ConA-treatment for observing hepatic injury and hepatocellular apoptosis. Results Comparison to ConA group,the mice in adiponectin group showed less hepatic injury and lower ALT level and less hepatocellular apoptosis(P 〈0.01), but those in leptin group showed a high hepatocellular apoptosis (P〈0.01) and high ALT levels. Conclusion Adiponectin may inhibit hepatocellular apoptosis and relieve hepatic inflammation, but leptin stimulats hepatocellular apoptosis and had no effection on ALT levels induced by ConA.
