摘要
目的观察异丙酚与依托咪酯脂肪乳剂预处理在大鼠短暂性脑缺血损伤时对海马神经细胞静息态钙离子(Ca2+)的影响,以进一步阐明它们的脑保护作用及其机制。方法健康雄性Wistar大鼠32只,随机分为假手术组(SH组)、缺血模型组(ISC组)、依托咪酯预处理组(EPC组)和异丙酚预处理组(PPC组),每组8只。应用激光共聚焦显微镜(LSCM)技术观察各组海马神经细胞内Ca2+荧光像素值的变化。结果大鼠脑缺血30 min后ISC组Ca2+荧光像素值较SH组显著增加(P<0.01),PPC组与EPC组较ISC组明显降低(P均<0.01),但两用药组间比较无显著性差异。结论异丙酚和依托咪酯预处理对大鼠短暂性脑缺血损伤的保护作用与抑制海马神经细胞Ca2+浓度的增加有关。
Objective It is to observe the effects of precondition of propofol and etomidate fat emulsion on cellular Ca^2+ ([ Ca^2+ ]i ) level in resting state in hippocampal neurons in rats with transient cerebral ischemia and to explore their cerebral protection and the mechanism. Methods 32 healthy male Wistar rats were randomly divided into sham operated group ( SH group), ischemia group (ISC group), etomidate pre-conditioning group (EPC group) and propofol pre-conditioning group (PPC group) (8 cases in each group). The fluorescence pixel value of [Ca^2+ ]i of each group was observed by the laser scanning confocal microscope. Results The fluorescence pixel value of [ Ca^2 + ]i in hippocampal neurons was markedly higher in ISC group than that in SH group (P〈0.01). And the values in both EPC group and PPC group were also lower than that in ISC group (P 〈 0.01 ), but there was no significant difference between EPC group and PPC group. Conclusion The protection of propofol and etomidate pretreatments for the transient cerebral isehemia injury in rats was associated with the inhibition of the increasing of [Ca^2+]i in hippocampal neurons.
