摘要
应用灭活嗜水气单胞菌作为凝集原,兔源抗血清作为凝集素,比较了在不同条件下微量凝集反应的结果。当菌液浓度约为108/ml,每孔反应液为30μl时,与常规的玻板凝集反应比较,可提高2~3个血清滴度。应用振荡器可加快反应,在10min内就可判读结果。温度(在10~37℃范围内)的影响不明显。应用该法和玻板凝集法同步检测免疫鱼鳖血清的结果表明,该法不仅具有敏感性高、提高检测率的优点(比平板法提高32%),而且还能定量检测鱼鳖血清抗体滴度和快速进行细菌鉴定(筛选鉴定了4株嗜水气单胞菌和6株水气单胞菌),为流行病学调查、免疫效果观察和细菌鉴定提供了敏感和可靠方法。
A micro-agglutination test was established with inactivated Aeromonas hydrophila (Ah) and rabbit anti-Ah-serum, and that the optimum reaction condition was investigated This method to measure serum antibody was more sensitive than slide agglutination test under the reaction condition with a bacterial density of 10 8/ml, and a total volume of antigen and antibody of reaction liquid of 30 μl/well, temperature range of 25~37℃ and reaction-time of 30 minutes, namely it could improve 2~3 titre and 32% of detection rate in comparison with the slide agglutionation In addition, as a quantitative measure method, it could be used to investigate epidemilogy of infection and to determine the immune effect after vaccination.
