eNOS基因治疗联合西地那非对大鼠急性低氧性肺动脉高压的预防作用

摘要 为探讨eNOS基因治疗联合西地那非对大鼠急性低氧性肺动脉高压的作用,将雄性Wistar大鼠36只随机分为常氧组(N)、低氧组(H)、低氧LacZ组(H-LacZ)、低氧eNOS组(H-eNOS)、低氧西地那非组(H-S)和低氧eNOS-西地那非组(H-eNOS-S),每组6只。首先经气道转基因(H-eNOS组和H-eNOS-S组应用5×109PFU/ml AdCMVceNOS600μl,H-LacZ组应用等量AdCMVLacZ,其它组应用等量生理盐水)。3天后,建立大鼠急性低氧性肺动脉高压模型,H-S和H-eNOS-S组大鼠在低氧前应用0.3%西地那非25 mg/kg,其它组大鼠应用等量生理盐水。低氧结束后测定大鼠mSAP和mPAP,并行肺组织eNOS免疫组化染色和cGMP、NO含量测定。结果显示:①H-eNOS组和H-eNOS-S组大鼠肺组织有广泛的eNOS转基因表达;②H-eNOS组和H-S组mPAP明显低于H组和H-LacZ组(P<0.01),明显高于H-eNOS-S组和N组(P<0.01),H-eNOS-S组和N组间无统计学差异(P>0.05);③各低氧组mSAP明显低于N组(P<0.05),且各低氧组间无统计学差异(P>0.05);④N组大鼠肺组织NO含量明显高于H组、H-LacZ组和H-S组,明显低于H-eNOS组和H-eNOS-S组(P<0.05),且H组、H-LacZ组和H-S组间以及H-eNOS组和H-eNOS-S组间无统计学差异(P>0.05);⑤H组和H-LacZ组大鼠肺组织cGMP含量明显低于N组(P<0.05),H-eNOS组和H-S组明显高于N组(P<0.05),且明显低于H-eNOS-S组(P<0.01)。结论:eNOS基因治疗联合西地那非,可协同性增加肺组织cGMP含量,具有选择性肺血管舒张作用,进而阻止大鼠急性低氧性肺动脉高压的发生。
