摘要
目的研究半边莲煎剂对肝癌H22荷瘤小鼠的抗肿瘤作用。方法建立肝癌H22荷瘤小鼠实体瘤模型,将24只小鼠分为3组:生理盐水组、半边莲煎组和5-氟脲嘧啶组各8只。通过测定荷瘤小鼠的瘤质量、抑瘤率观察半边莲煎剂对肿瘤生长的影响,采用免疫组织化学技术检测P27和Survivin的表达情况。结果半边莲煎剂能使肝癌H22荷瘤小鼠的瘤质量减少,抑瘤率为33.98%,显著低于对照组(P<0.05);P27和Survivin在3组中均呈阳性表达,单因素方差分析表明:半边莲煎剂组P27表达增强,Survivin表达减弱,与对照组相比差异均有统计学意义(P<0.05),半边莲煎剂组与5-Fu组比较,差异无统计学意义(P>0.05)。结论半边莲煎剂具有明显的抗肿瘤作用,其作用机制可能与P27和Survivin的表达有关。
Objective To study the anti-tumor effects of Chinese Lobelia Herb (CLH) decoction in H22 hepatocelluar carcinoma-healing mice. Methods H22 hepatocelluar carcinoma-bearing model was established in 24 mice which were then ramtomized to receive treatment with CLH decoction (CLH group) or 5-Fu (5-Fu group) compared to an untreated group (control group). The tumor-weights and rates of tumor inhibition were compared among the groups. P27 and Survivin in the tumor tissue were examined using immunohistochemical straining. Results CLH decoction was shown to reduce tumor weights in 39.48% of mice with significant difference compared to controls (P〈 0.05). Both P27 and survivin were detectable in the three groups. ANOVA revealed stronger expression of P27 and weaker expression of surviving in CLH group than in controls (P〈0.05) but no statistical difference between CLH and 5-Fu groups (P〉0.05). Conclusions The apparent anti-tumor actions of CLH may be related with its modulation on expressions of P27 and Survivin.
