摘要 本文研究了模板DNA浓度、镁离子浓度、引物浓度、dNTP及酶含量、退火温度及反应缓冲液中明胶或BSA含量等对ISSR反应体系的影响,通过优化反应条件初步建立了适合于剑麻ISSR反应的扩增体系,为进一步利用ISSR分子标记开展剑麻遗传多样性分析,种质资源鉴定和分子标记辅助育种等研究提供参考。 The main influencing factors of ISSR were studied and the experiment parameters were optimized. By adjusting the concentrations of DNA template, Mg^2+, dNTP,primer, the contents of Taq DNA polymerase, annealing temperature and PCR buffer (BSA or gelatin plus/free), a satisfactory ISSR reaction system for sisal was established. The results would provide reference for genetic diversity analysis, germplasm identification and marker-assisted selection of sisal with ISSR marker.
