组蛋白去乙酰化酶抑制剂的研究进展 被引量:3

摘要 核小体是真核生物染色质的基本单位,核小体的中部由4种组蛋白(H2A,H2B,H3,H4)各两分子形成八聚体,周围围绕DNA,尾部为组蛋白H1。核小体表面修饰与基因表达调控联系密切。组蛋白乙酰化是其中一种重要的共价修饰,通过招募染色体构型重建复合体引起染色体构型发生改变。染色体构型改变使高度螺旋的染色质变得相对松弛,便于转录因子与DNA结合。使组蛋白乙酰化的主要是组蛋白乙酰转移酶(histone acetyltransferase,HAT)和组蛋白去乙酰化酶抑制剂(histone deacetylase inhibitors,HDACI)。
