补气通阳颗粒对心肌缺血大鼠心血管活性物质释放的影响

The Effect of Buqitongyang Granule on the Release of Cardial and Vasoactive Substances in Rats with Myocardial Ischemia

目的探讨补气通阳颗粒对心肌缺血大鼠心脏和血管活性物质释放的影响。方法选取健康成年Wistar大鼠30只,随机分为空白对照组、模型对照组和模型治疗组,制作异丙基肾上腺素性心肌缺血动物模型,测定各组大鼠血浆中超氧化物歧化酶总活性(SOD)、血栓素B2(TXB2)、前列环素(PGI2)的中间代谢产物[TXB2、6-酮-前列腺素(6-Keto-PGF1α)]、血浆内皮素(ET)、血浆降钙素基因相关肽(CGRP)、心肌心钠素(ANF)以及心肌ATP酶活性等,并进行组间比较。结果与空白对照组比较,模型对照组大鼠SOD活性降低,TXB2、ET含量增加,6-Keto-PGF1α、CGRP减少,心肌ANF含量减少,ATP酶活性降低(P<0.05或P<0.01)。而模型治疗组改善情况优于模型对照组。结论补气通阳颗粒可通过提高SOD、ATP酶的活性以及纠正心肌缺血时血管活性物质释放的失衡而发挥对心肌和血管内皮细胞的保护作用。

Objective To investigate the effect of Buqitongyang granule on the release of cardial and vasoactive substances in rats with myocardial ischemia. Methods Thirty wistar rats were randomly divided into three groups：Normal control group（n=10） ,model control group（n= 10） and model treat group（n= 10） treated with Buqitongyang granule. The myocardial ischemia rat models were established with isopropylarterenol. Total activity of superoxide dismutase（SOD）,the contents of thromboxane B2 （TXB2） and 6 - keto- prostaglandin F1α（6- Keto- PGF1α ） （medial metabolism product of thromboxane A2 and prostaglandin I2 ）,the plasma level of endothelin （ET）, calcitionin gene related peptide（CGRP）, cardionatrin（ANF） and the activity of ATPase in myocardial were measured. Results Compared with normal control group,total activity of plasma SOD was lowered and the plasma levels of TXB2 and ET were increased in the model control group. The contents of ANF,CGRP and 6 -Keto -PGF1α were decreased in the model control group. The activity of myocardial ATPase were lowered in the model control group. All in the model treat group were obviously better than those in the control group and model control group,respectivily（P〈0.05 or P〈0.01）. Conclusion Buqitongyang granule had protective effect on myocardial and vessel endothlium, which might be related to enhancing the activity of ATPase and SOD and rectifying the release of vasoactive substances.
