夏天无对脑梗死循环内皮细胞和血清髓鞘碱性蛋白影响的实验研究 被引量:2

Experimental Study on the Effect of Rhizoma Corydalis Decumbentis on Circulating Endothelial Cells and Myelin Basic Protein in Cerebral Infarction Rats

摘要 目的观察夏天无对脑梗死模型大鼠循环内皮细胞(CEC)、血清髓鞘碱性蛋白(MBP)及脑组织病理形态学的影响,探讨其治疗脑梗死的可能机制。方法将60只Wistar大鼠随机分组,采用线栓大脑中动脉闭塞(MCAO)法制造脑梗死模型大鼠,用Percoll密度梯度分离法测定各组大鼠的血浆CEC数目,用ELISA双抗夹心法测定血清MBP及HE染色观测脑组织病理形态学的变化。结果夏天无高剂量组、低剂量组和曲克芦丁(维脑路通)组的CEC和MBP均较模型组降低(P<0.05或P<0.01),且高剂量组较低剂量组和维脑路通组降低更明显(P<0.05),同时,高剂量组、低剂量组和维脑路通组的脑组织病理形态学均较模型组有所改善,且高剂量组改善更明显。结论夏天无能降低脑梗死模型大鼠的CEC数量和血清MBP的浓度,同时能使脑梗死大鼠的脑组织病理形态学改善,起到保护脑血管内皮细胞和脑细胞的作用,这可能是其治疗脑梗死的机制之一。 Objective To observe the effect of Rhizoma Corydalis decumbentis on circulating endothelial cells(CEC),myelin basic protein(MBP) and the cerebral tissue pathomorphology in rats with cerebral infarction,and investigate its mechanisms. Methods Sixty wistar rats were randomly divided into groups. The middle cerebral artery occlusion( MCAO) rat models were eslablish by thread occlusion method. The changes of plasma CEC and serum MBP were observed by percoll density gradient separation and ELISA. The changes of cerebra, tissue pathomorphology were observed by hematoxylin and eosin staining. Results Compared with model group, the levels of plasma CEC and serum MBP were decreased in Rhizoma Corydalis decumbentis high dose group and Rhizoma Corydalis decumbentis low dose group and venoruton control group(P〈0.05 or P〈0.01). The levels of plasma CEC and serum MBPin Rhizoma Corydalis deeumbentis high dose group were lower than that in Rhizoma Corydalis decumbentis low dose group and venoruton control group(P〈0.05). The pathomorphology of cerebral tissue were improved in Rhizoma Corydalis decumbentis high dose group and Rhizoma Corydalis decumbentis low dose group and venoruton control group, and that in Rhizoma Corydalis decumbentis high dose group were more obvious. Conclusion Rhizoma Corydalis decumbentis can reduce the amount of plasma CEC and serum MBP concentrations, and improve the pathomorphology of cerebral tissue to protect cerebral vascular endothelial cell and brain cell in cerebral infarction rats,whtch may bca mechanism of treating cerebral infarction.
