摘要
目的观察大鼠矽肺形成过程中肺组织FIZZ1的动态变化及其与矽肺形成的关系。方法SD大鼠36只,随机分为2组,气管内注入二氧化硅混悬液复制实验性大鼠矽肺模型,采用苏木精-伊红染色、羟脯氨酸含量测定、免疫组织化学染色、实时荧光定量PCR等方法,观察实验性大鼠矽肺的发病过程及其肺组织中FIZZl蛋白、mRNA表达水平的动态变化。结果FIZZ1蛋白在正常肺组织为弱表达,实验性矽肺大鼠第7天时表达明显增强,30 d时较7 d时有所减弱,60 d时明显减弱,但仍强于对照组。FIZZ1 mRNA表达也显示出同样的结果。结论FIZZl蛋白和mRNA在实验性大鼠矽肺发病过程中呈现明显的动态变化,矽肺早期表达显著,在矽肺发生发展中可能起重要作用。
Objective To observe the dynamic changes of found in inflammatory zone 1(FIZZ1) in lung tissue of rats with experimental sillicosis and their correlation with sillicosis.Methods Thirty-six Sprague-Dawley rats were randomly divided into two groups.The rats in model group were injected with mixing liquid of silicon dioxide endotracheally,and those in blank control group with normal saline instead under the same conditions.By using HE staining of lung sections,determination of lung tissue hydroxyproline(HYP),immunohistochemical staining,real-time PCR,the changes in the expression of FIZZ1 protein and mRNA in lung tissue of rats with experimental silicosis were observed and analyzed.Results The expression of FIZZ1 protein in the lung tissue was increased at the 7th day in sillicosis animals,but slightly decreased on the 30th day,and expensively decreased on the 60th day,but stronger than in the blank control group.Real time-PCR revealed the same result.The expression levels of FIZZ1 protein and mRNA in sillicosis group were higher than in blank control group.Conclusion The expression of FIZZ1 protein and mRNA was dynamically changed in the lung tissue of rats with experimental sillicosis,especially in the early stage,suggesting FIZZ1 may play an important role in the sillicosis.
