一氧化氮及诱导型一氧化氮合酶与二硫化碳所致视网膜功能损害的相关性 被引量:1

Correlation between the Level of Nitric Oxide and Inducible Nitric Oxide Synthase in the Retina and Carbon Disulfide-induced Damage of Retina Function

摘要 目的探讨一氧化氮(NO)及诱导型一氧化氮合酶(iNOS)与二硫化碳(CS2)所致视网膜功能损害的关系。方法将48只SD雄性大鼠随机分为对照组、低剂量CS2染毒组和高剂量CS2染毒组。染毒结束后观察大鼠视网膜电图(ERG)b波振幅的变化;分光光度法测定NO含量;免疫组织化学法检测诱导型iNOS的表达。结果染毒组ERG的b波振幅均下降,视网膜iNOS表达增加,与对照组相比差异均有统计学意义,高剂量染毒组与低剂量染毒组之间差异亦有统计学意义。染毒组NO、iNOS分别与ERG的b波振幅呈负相关。结论NO及iNOS与CS2所致视网膜功能损害有关。 Objective To investigate the correlation between the level of nitric oxide(NO) and inducible nitric oxide synthase(iNOS) in the retina and carbon disulfide(CS2)-induced damage of retina function.Methods Forty-eight male SD rats were randomly divided into 3 groups: control group,group receiving high-dose CS2 and group receiving low-dose CS2.After treatment,amplitude of b-wave on electroretinogram(ERG) was observed.The content of NO was measured by spectrophotometer.The expression of iNOS was detected by immunohistochemistry.Results The amplitudes of b-wave on ERG were reduced in groups receiving CS2.The expression of iNOS and the contents of NO were increased in groups receiving CS2.There was significant difference between control group and groups receiving CS2,and there was also significant difference between group receiving high-dose CS2 and group receiving low-dose CS2.Negative correlation was found between the level of NO and iNOS in the retina and the amplitude of b-wave on ERG in groups receiving CS2.Conclusion NO and iNOS might be involved in CS2-induced damage of retina function.
