摘要
目的研究丁酸钠对人胃癌细胞系生长抑制及诱导分化的抗肿瘤作用。方法以不同浓度的丁酸钠作用体外培养的人胃癌细胞株SGC-7901,应用MTT法检测对细胞生长的抑制情况;应用分光光度法检测分化标志酶活力;应用流式细胞技术检测细胞周期分布。结果丁酸钠可以使SGC-7901细胞发生形态学改变;可抑制细胞生长,2.5mmol/L和5.0 mmol/L两种浓度丁酸钠在24、48、72 h对细胞生长的抑制率分别为27.72%、39.25%、85.02%和39.05%、60.76%、93.63%,具有时间剂量依赖性(P<0.01);可使细胞分化标志酶乳酸脱氢酶(LDH)和碱性磷酸酶(ALP)活力降低;可使G0/G1期细胞大量增多,S期、G2/M期细胞相应减少,出现G0/G1期停滞。结论丁酸钠可诱导SGC-7901细胞分化并抑制其恶性生长。
Objective To investigate the antitumor effect of sodium butyrate(NaB) on human gastric carcinoma cells.Methods SGC-7901 cells cultured in RPMI-1640 were treated with NaB at different concentrations.Morphological changes of SGC-7901 cells were observed under an inverted microscope.MTT assay,flow cytometry,and spectrophotometry were performed to determine the cell proliferating activity,cell cycle distribution,and enzyme activity,respectively.Results After treated with NaB,SGC-7901 cells showed morphologic changes and significant inhibition of growth.The inhibition rate of growth of cells treated with NaB at concentrations of 2.5,and 5.0 mmol/L were 27.72% and 39.05%(24 h),39.25% and 60.76%(48 h),and 85.02% and 93.63%(72 h) respectively,and the inhibitory effects were time-and dose-dependent(P〈0.01).The activities of LDH,and ALP were suppressed significantly(P〈0.01).The number of cells in G0/G1 phase was increased massively,that in S and G2/M phase decreased correspondingly(P〈001),and cells were arrested in G0/G1 phase.Conclusion NaB can induce differentiation of SGC-7901 cells,and suppress their malignant growth.
