摘要
目的:建立RP-HPLC法同时测定三黄片中黄芩苷、小檗碱、大黄素和大黄酚的含量。方法:色谱柱为KromasilKR100-5 C18柱(250 mm×4.6 mm,5μm);流动相:乙腈(A)∶3%乙酸水溶液(B);梯度洗脱程序:A(%)∶B(%)=23∶77(0-9 min),流速1 mL/min,A(%)∶B(%)=28∶72(10-21 min),流速0.7 mL/min,A(%)∶B(%)=66∶34(22-35 min),流速1 mL/min;检测波长270 nm;柱温35℃。结果:四组分均达到基线分离,各组分的平均回收率为97.00%-102.55%,方法的精密度、稳定性和重现性良好。结论:本方法快速、可靠、简便,具有应用价值。
Objective:To establish a RP-HPLC method for simultaneous determination of contents of four components in Sanhuang tablets.Methods:HPLC was used to detect the contents of the four components:baicalin,berberine,emodim and chrysophanol.The study was carried out on a Kromasil KR100-5 C18 column(250 mm×4.6 mm,5 μm) with acetonitrile(A)-3% acetic acid water solution(B) gradient elution system as mobile phase.A(%)∶B(%) was 23∶77 in 0-9 min with flow rate at 1 mL/min,A(%)∶B(%)=28∶72 in 10-21 min with flow rate at 0.7 mL/min and A(%)∶B(%)=66∶34 in 22-35 min with flow rate at 1 mL/min.The detective wavelength was 270 nm and the column temperature was 35 ℃.Results:The four components were separated on the baseline.The average recovery rates were in the range of 97.00%-102.55%.The accuracy,repeatability and stability of the method were good.Conclusion:This method is rapid,reliable,simple and applicable.
