高效液相色谱-蒸发光散射法测定黑归脾丸中黄芪甲苷含量

Determination of Astragaloside in Heiguipi Pills by HPLC-ELSD

目的建立黑归脾丸的定量控制方法。方法采用高效液相色谱-蒸发光散射(HPLC-ELSD)法测定黑归脾丸中黄芪甲苷的含量,色谱柱为Zorbax SB-C18柱(250mm×4.6mm,4.6μm),流动相为甲醇-水(75∶25),流速0.5mL/min,柱温30℃,蒸发光散射检测器的漂移管温度为90℃,气体流速为1.5L/min。结果黄芪甲苷进样量在0.428～10.7μg内与峰面积线性关系良好,平均回收率为100.80%,RSD=5.38%(n=6)。结论HPLC-ELSD法简便、重现性好,可用于控制黑归脾丸的质量。

Objective To establish a method of the quantitative determination of Heiguipi Pills. Methods The content of astragaloside was determined by HPLC-ELSD. Chromatographic conditions were Agilent Zorbax SB-C18 column （250 mm×4.6 mm,4.6 μm）,the mobile phase was methanol- water（75 ： 25）,the flow rate was 0. 5 mL/min with column temperature at 30℃, the drifttube temperature of ELSD was 90 %, and the flow rate of air was 1.5 L/min. Results The good linearity was observed for astragaloside in the range of 0. 428 - 10. 7 μg. The average recovery rate of astragaloside was 100.76% with RSD was 5.38% （n = 6）. Conclusion HPLC- ELSD is simple and accurate with good repeatability. It can be used for the quality control of Heiguipi Pills.
