Syk和SYK-mRNA在乳腺浸润性导管癌中的表达及其与腋窝淋巴结转移的关系

摘要 目的 探讨Syk及其编码基因SYK—mRNA在乳腺癌中的表达及其与腋窝淋巴结转移的相关性。方法分别采用免疫组化SP法和原位RT—PCR方法同步检测52例乳腺浸润性导管癌(其中有腋窝淋巴结转移者32例)和39例乳腺非癌组织中Syk蛋白和SYK—mRNA的表达。结果Syk和SYK—mRNA在乳腺癌中的阳性表达率分别为42.3%(22/52)和38%(20/52),均显著低于非癌组织。Syk和SYK—mRNA表达的一致率为91%。无淋巴结转移组的乳腺癌Syk和SYK—mRNA阳性表达率均显著高于有淋巴结转移组的乳腺癌,但两组的Syk和SYK—mRNA表达量差异均无统计学意义。结论Syk(SYK—mRNA)的表达缺失与乳腺癌腋窝淋巴结转移有关,SYK—mRNA基因可能是乳腺癌的一种候选抑癌基因。 Objective To investigate the expression of Syk ( SYK - mRNA) and its relationship with axillary lymph node metastasis in breast invasive ductal carcinoma. Methods The expression of Syk protein and mRNA was detected by immunohistochemical SP method and in situ PCR respectively in 52 cases of breast invasive ductal carcinoma( 32 cases with metastatic axillary lymph nodes )and 39 noncancerous breast tissues. Results The positive expression rate of Syk protein and mRNA was 42.3% (22/52) and 38% (20/52)respectively in the tumor tissues ,which was significantly lower than in the non - cancerous tissues ( X^2 = 3 1. 01, P 〈 0.05, X^2 = 27.94, P 〈 0.05 ). The concordance rate of Syk protein and mRNA was 91% ( X^2 = 57.50, P 〈 0. 05 ; kappa = 0. 815, P 〈 0.05 ). The positive expression rate of Syk protein and mRNA in the group with non - metastatic axillary lymph nodes was significantly higher than in the group with metastatic axillary lymph nodes ( X^2 = 14.23, P 〈 0.05 ; X^2 = 6.37, P 〈 0.05 ), but there was no statistically significant difference in the Syk protein and mRNA expression levels between the two groups(t =0.749,P =0.46;t = -0.48,P =0.632). Conclusion Reduction or loss of Syk protein and mRNA expression may be correlated to lymph nodes metastasis of breast invasive ductal carcinoma, indicating that SYK gene may be a candidate tumor suppressor of breast cancer.
