乙肝病毒X蛋白对E—cadherin和T—cadherin表达的调节作用 被引量:1

摘要 目的 体外研究乙肝病毒X蛋白(HBx)对E-钙黏蛋白(E—cadherin)和T-钙黏蛋白(T—cadherin)表达的调节作用。方法建立稳定表达HBx基因的HepG2细胞系(HepG2-HBx),稳定转染空质粒pcDNA3.1的HepG2细胞系作为对照(HepG2-pcDNA3.1),分别用荧光实时定量(Real—time)PCR和Western—blot技术检测转染前后E—cadherin和T—cadherin在mRNA及蛋白水平表达变化。结果以HepG2-pcDNA3.1细胞为参照,转染HBx基因后的HepG2-HBx细胞E—cadherinmRNA和蛋白水平分别为其(42.28±8.73)%和(35.28±4.33)%,而T—cadher—in则分别为其(4.47-4-0.78)和(5.11±0.83)倍,差异有统计学意义(P〈0.05)。结论HBx下调E—cadherin的表达,上调T—cadherin的表达,其机制可能为HBx促进E—eadherin向T—cadherin转换。 Objective To study the effect of hepatitis B virus X protein (HBx) on the expression of E - cadherin and T - cadherin in vitro. Methods HepG2 - HBx cell line with stable expression of HBx gene was established, and HepG2 - pcDNA3. 1 cell line transfected with empty plasmid served as a control. The mRNA and protein levels of E - cadherin and T - cadherin expression were detected by real - time PCR and Western blot. Results As compared with control cells, the mRNA and protein levels of E - cadherin in HepG2 - HBx cells were decreased [ ( 42.28 ± 8.73 ) % and ( 35.28 ± 4.33 )% respectively] , but the mRNA and protein levels of T- cadherin in HepG2 -HBx cells were increased [ ( 4.47 ± 0.78 ) and ( 5.11 ±0. 83 ) folds respectively ]. The differences were of statistical significance( P 〈 0.05 ). Conclusion HBx down - regulated the expression of E - cadherin, and up - regulated the expression of T - cadherin. HBx may play an important role in promoting the caherin switch from E - cadhefin to T - cadherin.
