摘要
以文心兰茎尖为材料,研究生长调节剂、碳源等因子对原球茎诱导与植株再生的影响。结果表明:0.1mg/LTDZ生长调节剂适合原球茎诱导,且无不定芽分化,诱导率高达80%;果糖为原球茎增殖最佳碳源,其浓度以10g/L效果较好,增殖系数达8.15;不同浓度的BA,TDZ和2ip比较发现,1.0mg/L2ip对原球茎分化效果最好,平均每克原球茎分化35.7个芽;0.5mg/LIBA与0.2mg/LNAA配合使用生根效果较好,生根率达100%,平均生根数为3.8条,根长可达23.0mm。
Oncidium 'Gower Ramsey' (Oncidium luridum Ldl. cv Gower Ramsey) was cultured with shoot tips as materials on the medium supplemented with different hormones and carbon sources to induce protocorm-like bodies(PLBs) for subsequent plant regeneration. The results showed that 0.1 mg/L of TDZ was more effective in inducing PLBs without shoots with an inducing rate of 80%. Fructose was the suitable carbon source with the better result for the proliferation of PLBs at the concentration of 10 g/L, and the proliferation coefficient was 8.15. The 2ip was a more efficient cytokinin to induce bud differentiation than BA and TDZ, and induced 35.7 buds per gram of PLBs at a concentration of 1.0 mg/L. Rooting medium supplemented with a combination of 0.5 mg/L IBA and 0.2 mg/L NAA promoted rooting of plantlets with a 100% rooting rate, and each shoot produced 3.8 roots which were 23.0 mm long by average.
