白银豆2个内参基因片段的克隆与初步分析 被引量:1

Cloning and Analysis of Two Internal Control Gene Fragments from Phaseolus lunatus L.

摘要 以白银豆幼苗为研究对象,克隆2个常用候选看家基因肌动蛋白基因(actin)和18SrRNA的片段,并应用半定量RT-PCR技术分析它们在不同组织及低温处理叶片中的表达。结果表明,各样本间的表达量无显著差异,说明actin和18SrRNA可用于校正白银豆目标基因的表达量。 Quantitative data of target gene are often normalized by transcription levels of reference or housekeeping genes in gene expression experiments. Two frequently used housekeeping gene fragments namely actin and 18S rRNA were cloned from Phaseolus lunatus L., and their expression levels were assessed by semi-quantitative RT-PCR in 3 tissues(root, stem and leaf) and in the leaves treated with low temperature. The results showed no significant changes in expression among the tested samples. Both actin and 18S rRNA can be therefore used as internal controls for gene expression analyses in Phaseolus lunatus L.
