利用ISSR标记鉴别珍珠黄杨无性系 被引量:1

摘要 利用ISSR-PCR方法对珍珠黄杨5个无性系进行了基因组多态性分析,从100个ISSR引物中筛选出19个多态性引物用于扩增,共扩增出256条谱带,其中多态性条带211条,占总带数的82.4%,平均每条引物扩增的DNA带数目为13.5条,依据扩增结果进行Ne i相似性系数和遗传距离分析,利用UPGMA法构建聚类树状图。结果表明:ISSR分析产生了一些无性系特有的指纹图谱,认为ISSR技术在珍珠黄杨无性系鉴定和阐明遗传关系中有一定的应用潜力。 Inter-simple sequence repeats (ISSR) was used to determine the genomic DNA variations in five clones of Buxus sinica var. parvifolia in this paper. Nineteen polymorphic primers were screened from 100 primers, there were 256 reproducible and clear amplification products, 211 of which were polymorphic (82.4%), the average number of DNA bands amplified by each primer was 13.5. By the Nei's genetic distance and UPGMA cluster analysis, we found the potential diagnostic fingerprints for various clones, ISSR marker was an efficient method for cultivar identification.
