摘要
采用RT-PCR法从水稻中成功克隆了1个水杨酸甲酯转移酶基因(OsSAMT1),构建了该基因的超表达载体,通过农杆菌介导法转化粳稻品种日本晴,获得10个转基因阳性植株.生物信息学分析结果表明,水稻基因组中共有12个OsSAMT1的同源基因,它们分别属于第6和第11号染色体上的2个基因族,而位于第2号染色体上的OsSAMT1基因与第6号染色体上的同源基因具有更高的相似性.
As OsSAMT1, a MeSA transferase gene, was successfully separated and cloned from rice by means of RT-PCR strategy, overexpression construct was prepared and ten positive transgenic plants were obtained with Agrobacteriummediated method. Bioinformatics analyses show that there are twelve homologous genes of OsSAMT1 in rice genome, they mainly belong to two gene families on chromosome six and eleven respectively, and OsSAMT1 shows higher similarity with those on chromosome six. These results paved a solid way for deep functional study of OsSAMT1 gene, especially in disease resistance molecular mechanism in plant.
