三七总皂苷在抗肾纤维化细胞模型中剂量的研究 被引量:5

摘要 目的研究三七总皂苷(PNS)在抗肾纤维化细胞模型中剂量选择。方法原代培养大鼠肾小球系膜细胞(MC)作为细胞模型,用含有或者不含有胎牛血清的各浓度(0.05,0.1,0.2,0.4,0.6,0.8,1.0,1.2,1.6,3.2mg/mL)三七总皂苷细胞培养刺激大鼠MC,MTT比色法观察其对细胞的增殖及毒性作用。结果PNS浓度在0.1~0.6 mg/mL抑制大鼠MC细胞增殖(P〈0.05),但对大鼠MC不具有细胞毒性作用(P〉0.05),而PNS浓度≥0.8 mg/mL时具有细胞毒性作用(P〈0.05),〈0.1 mg/mL时对MC没有抑制增殖的作用。PNS浓度在0.1~3.2 mg/mL与大鼠MC增殖呈负相关(r=-0.838,P〈0.01)。结论浓度0.1~0.6 mg/mL是PNS在抗肾纤维化细胞模型中理想剂量范围;在该范围内PNS抑制MC增殖的作用不是通过细胞毒性实现的。
