抗rhG--csf单克隆抗体相对亲和力分析

DETERMINATION OF RELATIVE AFFINITY OF MONOCLONAL ANTIBODIES AGAINST rhG csf

目的：检测鼠抗人Ｇ－ｃｓｆ单克隆抗体的相对亲和力。方法：采用间接酶联免疫吸附试验（ＥＬＩＳＡ）。结果：各株单抗结合抗原的相对亲和力不同，根据５０％最大结合浓度分为２组，其中４株杂交瘤上清ＭｃＡｂｓ与纯化腹水ＭｃＡｂｓ测得结果基本吻合。结论：实验结果为合理运用这些单抗提供了重要依据。

Objective:To measure the relative affinity of anti G csf monoclonal antibodies.Method:Indirect enzyme linked immunosorbent assay (ELISA) was used.Results:The affinity of various McAbs to the same antigen was different and some of the difference was very evident.Six McAbs were divided into two groups by analyzing their 50% maximum binding.The relative affinity of McAbs in the culture supernate was consistent with that in the purified ascites.Conclusion:The study provides useful information for the rational application of these McAbs.
