旋毛虫新生幼虫抗原免疫小鼠免疫功能的动态观察

摘要 【目的】探讨旋毛虫新生幼虫抗原诱导小鼠产生的保护性免疫机制。【方法】采用免疫组化法分别检测旋毛虫新生幼虫抗原免疫后7、14、28、42 d小鼠外周血CD4+和CD8+T淋巴细胞的百分率,并同时用酶联免疫吸附试验(ELISA)测定外周血PBMC培养上清中IL-4和IFN-γ水平。【结果】与对照组相比,旋毛虫新生幼虫抗原免疫组小鼠外周血CD4+、CD8+T淋巴细胞的百分率均增高,IL-4和IFN-γ水平亦升高,直至免疫后42 d;而CD4+/CD8+T细胞比值与对照组相比无显著性差异(P>0.05)。【结论】旋毛虫新生幼虫抗原能诱导机体产生有效的体液免疫和细胞免疫,有希望成为预防旋毛虫病有前途的候选疫苗。 [Objective] To study the immunoprotective mechanism of mice immunized with antigens derived from new- born larvae of Trichinella spiralis. [ Methods] The percentages of CD4^+ and CD8^+ T cells in peripheral blood were examined by immunohistochemical method on 7, 14, 28, 42 days after mice were immunized with antigens of newborn larvae of Trichinella spiralis.At the same time, peripheral blood mononuclear cells (PBMC) were isolated and cultured. The supernatants were collected and the levels of IL-4 and IFN-γ were detected by enzyme-linked immunosorbent assay (ELISA). [Results] Compared with control group, the percentages of both CD4^+ T cell and CD8^+ T cell in peripheral blood increased, meanwhile the levels of IL-4 and IFN-γ also increased in newborn larva antigen group, which kept to the 42 days after immunization. The ratio of CD4^+/ CD8^+ T cells was not significantly different in newborn larva antigen group as compared with that of control group ( P 〉 0.05). [Conclusions] Newborn larva antigens of Trichinella spiralis can stimulate the mice to produce obvious protective humoral immunity and cellular immunity and they are very likely to become a candidate of promising vaccine.
