摘要
Exocyst复合物是由SEC-3、SEC-5、SEC-6、SEC-8、SEC-10、SEC-15、EXO-70和EXO-84共8种蛋白亚基组成的重要囊泡栓系因子.尽管已经被广泛地研究,但是对于该复合物的认识仍然有限.通过基因敲除的方法,获得了Sec-10基因突变的线虫,并发现该线虫存在产卵障碍的表型.在对性腺形态、精子功能、卵黄吸收、AC细胞的融合以及神经对产卵区域肌肉控制等产卵途径中若干关键因素的进一步研究中,发现Sec-10基因突变的线虫与野生型相比不存在明显区别.因此需要进一步的研究来阐明该表型产生的原因.
Exocyst complex plays an important role in the organisms. It includes SEC-3, SEC-5, SEC-6, SEC-8, SEC-10, SEC-15, EXO-70 and EXO-84. Although it has been studied broadly, there are still a lot of mechanisms that need to be uncovered. In order to know this complex more clearly, we obtained a Sec-10 mutant strain using knockout method. The Sec-10 mutant strain showed an abnormal egg-laying phenotype. After studying several key factors involved in the egg-laying process, including gonad formation,sperm function, yolk endocytosis, the fusion between AC and utse, HSNs and VCs, we found no obvious differences between Sec-10 mutant and wild-type strain. Further work needs to be done to explore the mechanism underlying the egg- laying phenotype of Sec-10 mutant strain.
