香菇多糖脂质体与注射剂对正常小鼠免疫效应的比较

Comparison of the immune regulatory effect of the lentinan liposome and injection in normal mice

目的:比较香菇多糖脂质体与注射剂对正常小鼠免疫功能影响,评价香菇多糖引入脂质体剂型的免疫效应。方法:NIH小鼠随机分到香菇多糖脂质体0.5,1.0,3.0mg.kg-13个剂量组,注射用香菇多糖1mg.kg-1组,对照组给予等量葡萄糖注射液,腹腔注射给药,qd,连续10d。给药结束后依据廓清率实验,体内淋巴细胞转化实验和血清溶血素实验方法的观测指标,评价不同剂型香菇多糖对小鼠巨嗜细胞吞噬能力,T淋巴细胞转化率及体液免疫功能的影响。结果:香菇多糖脂质体可以增强巨嗜细胞吞噬能力,促进外周血和脾脏T淋巴细胞增殖,提高动物体液免疫功能,其效应与剂量相关。吞噬指数分别为(3.9±0.9),(4.2±0.8),(4.7±1.0);脾脏T淋巴细胞转化率分别为(50.6±8.8)%,(60.0±5.2)%和(63.4±9.1)%;血清溶血素分别为(1640.9±380.1),(2002.0±448.6),(1901.2±369.8)。香菇多糖注射剂亦能提高动物的免疫功能,与同剂量香菇多糖脂质体相比,其脾淋巴细胞转化能力和体液免疫功能弱于脂质体,差异具有显著性。结论:香菇多糖引入脂质体制剂后,依然保留了增强巨嗜细胞吞噬能力,促进外周血和脾脏T淋巴细胞增殖,提高动物体液免疫的功能,与注射剂相比其效应有所提高。

OBJECTIVE To compare the immune regulatory effect of the lentinan liposome and injection in normal mice. METHODS The experimental mouse was injected with high,middle,and low dosages of lentinan liposome （0. 5,1,3 mg·kg ^-1 ）in the abdomen（i, p. ） ,Lentinan （1 mg·kg^- 1·d ^-1 ）and the comparison group was given with the same volume of glucose i. p. once a day. After treatment for 10 consecutive days, carbon granular-clearance, the weight ofimmune organ, the ability of the spleen lymphocytes proliferation and the hemolysinproduction test was used to evaluate the influence of Lentinan liposome on immunity in normal mice. RESULTS The lentinan liposome could promote the carbon granular clearantce,accelerate the proliferation of T lymphocyte in blood and spleen, enhance the hemolysin production, and the effect is correlated with dosage. The len- tinan injection still could improve the immune regulatory effect, but it was weeker than liposome statistically. CONCLUSION The lentinan liposome has certain potential effect on the phagocytic function, T lymphocyte and immunity regulation,even bet ter than injection.
