摘要
目的:了解大鼠脑出血后病灶周围神经干细胞增殖情况。方法:将大鼠随机分为:单纯脑出血组20只,3、7、14、28、60天各4只,生理盐水组10只,3、7、14、28、60天各2只。应用立体定向技术,单纯脑出血组用大鼠自体尾动脉不抗凝血液75μl缓慢注入大鼠尾状核,制成中等量脑出血模型,生理盐水组将等量生理盐水缓慢注入大鼠尾状核,模型制作后笫2天开始腹腔注射5-溴尿嘧啶(BrdU)每天2次,共10次。分别在术后3、7、14、28、60天灌注动物,用BrdU免疫荧光和Hoechst标记细胞核,观察BrdU阳性神经干细胞的增殖情况。结果:生理盐水组未发现神经干细胞的增殖,脑出血组在3、7天未发现增殖,14、28、60天组发现少量的神经干细胞。结论:脑出血后第14、28、60天在基底节脑出血病灶周围可出现神经干细胞增殖。
Objective:To explore the proliferation of endogenous neural stem cell(NSCs) following cerebral hemorrhage.Methods:The rats were divided randomly as follows:the simple hemorrhage group,including 20 rats,with 4 rats in each subgroup of 3,7,14,28 and 60d;the normal saline(NS) group,including 10 rats,with 2 rats in each subgroup of 3,7,14,28 and 60d.Stereotaxic technique was used.75μl coagulated blood was injected into the rat caudate nucleus to make the medium cerebral observe hemorrhage model.The samle amount of NS was injected into the rat caudate nucleus. BrdU was was injected into abdomen tid day,all ten time. The rats in both groups were raised for different time to observe the proliferation of NSCs. The rats were perfused at 3,7,14,28 and 60d after op- eration. The nuclei of endogenous neural stem cells (NSCs) were showed by immunofluorescence and Hoechst. The immunochemistry was used to observe proliferation of endogenous NSCs. Results: There was no proliferation of NSCs found in the the normal saline (NS) group and 3d,7d in simple hemorrhage group.While small numbers of endogenous NSCs were found in the 14d, 28d, 60d subgroups in the simple hemorrhage group. Conclusion: some endogenous NSCs are found in 14d,28d and 60d after the cerebral hemorrhage.
