摘要
目的通过培养的大鼠脑微血管内皮细胞(BMVEC),研究不同浓度的同型半胱氨酸(Hcy)对内皮细胞损伤的影响。方法将培养的BMVEC分成两大组,将其与不同浓度的Hcy(50、200μmol/L和1、2 mmol/L)和(或)无CuSO4(4μmol/L)共同培养18 h,分别利用噻唑蓝、细胞苏木精-伊红染色和流式细胞仪技术观察BMVEC细胞的生长及凋亡情况。结果BMVEC在CuSO4的协同下与不同浓度的Hcy培养18 h后,细胞活力A值呈现剂量依赖性的降低,超过1 mmol/L的Hcy对BMVEC已经形成了显著的抑制和毒性效应,滞留于G1期的细胞比例增加。结论高浓度的Hcy具有内皮细胞毒作用,可以抑制细胞的生长。
Objective To study the changes of endothelial dysfunction in cultured brain micro-vascular endothelial cells(BMVECs)with Hcy in different concentrations.Methods BMVECs were divided into 2 big groups:Hcy with or without CuSO4(4 μmol/L).And the Hcy was used in different concentrations(50 μmol/L,200 μmol/L,1 mmol/L、2 mmol/L).By means of MTT、HE and FCM to observe the cell cycle and endothelial dysfunction after the BMVEC were cultured for 18 hours.Results After BMVECs were exposed to 4 μmol/L copper plus Hcy in different concentrations for 18 hours,the survival rate was decreased significantly in a dose-dependent manner;and the rates of arrested cells in G1 was increased.Conclusion Hcy could lead to ultrastructural damage of endothelial cell.
