摘要
本研究以5个春麦品种的幼穗为外植体,接种在MS为基本培养基不同浓度的激素的诱导培养基上,结果发现,铁春2号是最好的基因型,诱导率、分化率最高,不同品种间诱导率存在极显著差异,6-BA不利于愈伤组织诱导,最佳生长素配比是20mgL-12,4-D+05mgL-1NAA。在分化培养中,最好的激素配比为02mgL-1NAA+01mgL-1Kt。经长期继代的愈伤组织5℃低温处理10d,显著提高愈伤组织的分化率。
The explants of the young panicles of 5 spring wheat varieties were cultured in induced MS The result showed that the variety Tiechun 2 was the best genotype and its induction and differentiation frequency were the highest among 5 varieties Difference of induction frequency was significant between wheat varieties 6-BA had negative effect on callus induction. The best combination of growth horomone for callus induction was 2.0 mg.L -1 2,4-D+0 5mg L -1 NAA. The best combination of growth horomone for differentiation was 0 2mg.L -1 NAA+0 1mg.L -1 Kt. Differentiation frequency of callus was significantly high under 5℃ with 10 days in long period subculture.
