猪传染性胃肠炎病毒N蛋白的二级结构和B细胞抗原表位的预*测

摘要 以TGEV基因组序列为基础,运用Garnier—Robson、Chou—fasman和Karplus—Schulz方法预测猪传染性胃肠炎病毒N蛋白的二级结构。运用Kyte—Doolitte方法对N蛋白的疏水性进行了分析,Jameson-Wolf方法预测了结构蛋白的抗原指数,Emini方法预测了N蛋白的表面可能性,最后结合吴玉章的方法综合分析预测B细胞抗原表住。结果显示柔性区域主要集中在N-端第11~31、52~65、128~147、155~188、210~227、313~319、331~347区段,α-螺旋和β-折叠数量少且分散。N蛋白可能的B细胞抗原位点是N-端12~31,154~191,204~243,309~320,329~354区段,而这些区段内部或附近都有柔性区域存在,可作为N蛋白的抗原优势表位。本研究为TGEV抗原优势表位的试验研究和反向疫苗的研制奠定了基础。 To predict the secondary structure of TGEV nucleoprotein with the methods of GarnierRobson.Chou-fasman and arplus-Schulz. Hydrophilicity , surface probability plot and antigenic index were obtained by the methods of Kyte-Doolittle, Emini and Jameson-Wolf respectively, then integrated the method of WU Yu-zhang to analysis B cell epitops. The results showed that the flexible regions at the N- terminal No. 11-31,52-65,128-147,155-188,210-227,313-319,331-347, a few α-helix and β- sheet regions were distributed in the protein. B cell epitops of TGEV nucleoprotein were probably located at or adjacent the N-terminal No. 12-31, 154-191, 204-243,309-320,329-354 regions, there are flexible regions in or hereabout these regions, they can be used as the preponderant epitops of nucleoprotein. This article established the basis for the examination and monoclonal antibody of the preponderant epitops of TGEV nucleoprotein.
