摘要
利用PCR-RFLP鉴定体系和SDS-PAGE蛋白分析方法,分析了从我国千山地区不同森林地带土壤中分离的10株Bt菌的32类杀虫晶体蛋白基因类型和表达蛋白。研究表明,电镜照片显示晶体形状较为多样,有长菱形、菱形、方形、球形。同时含有cry1、cry7、cry16类基因的有1株菌,同时含有cry1、cry2类基因的有1株菌,含有cry30类基因的有1株菌,含有cry1基因的有3株菌,4株菌不含所鉴定的32类基因。同时也发现,绝大多数含有cry1基因的菌株表达了130ku蛋白,含有cry2基因的菌株表达了60ku的蛋白,含有cry30基因的菌株表达了30ku的蛋白。
Based on the methods of PCR-RFLP and SDS-PACE, cry genes and their expression products of 10 Bacillus thuringiensis isolates from different forestry site zones in Qianshan of China were analyzed. The ICPs showed that many different shapes were observed by electric microscope including the diamond crystal, square crystal, and sphericity crystal. The results showed that one strain contained three cry-type genes: cry1, cry7 and cry16, one strain contained both cry1 and cry2 genes, one strain contained cry30 type genes, three strains contained cry1 type gene, four strains did not contain anyone of 32 cry-type genes. The results also showed that most of the cry1 genes encoded 130 ku proteins, cry/2 genes encoded 60 ku proteins, and cry30 genes encoded 30 ku proteins.
