黄芪多糖对大鼠肌组织能量代谢的影响 被引量:9

摘要 目的观察黄芪多糖(APS)对游泳大鼠糖代谢相关酶的影响,探讨APS对大鼠能量代谢影响的作用机制。方法采用实验法检测腹腔注射APS大鼠(APS组)游泳力竭时间和不同状态下骨骼肌琥珀酸脱氢酶(SDH)、苹果酸脱氢酶(MDH)、乳酸脱氢酶(LDH)活性以及乳酸水平的动态变化,并与腹腔注射等量0.9%氯化钠溶液的大鼠(对照组)进行比较。结果(1)对照组(24只)和APS组(24只)大鼠游泳力竭时间分别为(6.1±1.8)h和(9.3±2.9)h,差异有统计学意义(t=2.35,P<0.05)。(2)对照组大鼠安静状态下骨骼肌SDH活性(12.2±2.8)U/mgprot、MDH活性(0.56±0.05)U/mgprot及力竭即刻骨骼肌乳酸水平(0.74±0.21)mmol/gprot与ASP组分别为(15.7±2.9)U/mgprot、(0.77±0.10)U/mgprot和(0.42±0.09)mmol/gprot比较,差异有统计学意义(P<0.05)。(3)对照组大鼠游泳力竭即刻SDH活性(9.3±1.1)U/mgprot、运动1h时MDH活性(0.68±0.11)U/mgprot、力竭即刻MDH活性(0.44±0.04)U/mgprot、力竭即刻LDH活性(191±22)U/100gprot与安静状态下SDH、MDH、LDH活性分别为(12.2±2.8)U/mgprot、(0.56±0.05)U/mgprot和(258±50)U/100gprot比较,差异均有统计学意义(P<0.05)。结论APS可以提高大鼠机体有氧代谢酶SDH、MDH的活性,使能源物质得到充分利用的同时减少了对机体有危害的代谢产物的生成,从而维持更长时间的运动;APS对骨骼肌LDH活性、LD水平影响不明显,但有利于代谢产物乳酸的清除,从而可以延缓疲劳的产生。 Objective To observe the effects of astragalus polysaccharide(APS) on the enzyme related to glucose metabolism of skeletal muscle in swimming rat,to explore the mechanism of action of APS influencing rat energy metabolism.Methods Experimentation was used to determine the swimming power exhaustive time and the dynamic changes of skeletal muscle succinate dehydrogenase(SDH),malate dehydrogenase(MDH) activity and lactate dehydrogenase(LDH) in rats injected with APS(APS group).The indexes were compared with that of rats injected with equal 0.9% sodium chloride solution(control group).Results The swimming power exhaustive time of groups control(24 rats) and APS(24 rats) was (6.1±1.8)h,(9.3±2.9)h,respectively,the difference was significant(t=2.35,P〈0.05).There was significant difference between groups control and ASP in skeletal muscle SDH activity(12.2±2.8 U/mgprot,15.7±2.9 U/mgprot,respectively),MDH activity(0.56±0.05 U/mgprot,0.77±0.10 U/mgprot,respectively) in a quiet state and lactic acid content instantly after power exhaust(0.74±0.21 mmol/gprot,0.42±0.09 mmol/gprot,respectively).In control group,SDH activity instantly after swimming power exhaust,MDH activity after 1 h swimming or instantly after power exhaust,and LDH activity instantly after power exhaust were (9.3±1.1)U/mgprot,(0.68±0.11)U/mgprot,(0.44±0.04)U/mgprot,(191±22)U/100 gprot,respectively,and SDH,MDH and LDH activities in a quiet state were (12.2±2.8)U/mgprot,(0.56±0.05)U/mgprot,(258±50)U/100gprot,respectively,the difference was significant(P〈0.05).Conclusion APS can improve the activities of oxygen-metabolizing enzymes SDH,MDH,and reduce the production of metabolic products harmful to body while making full use of energy materials,thus to maintain a longer movement.The effect of APS on skeletal muscle LDH activity and LD level was not obvious,but conducive to removal of lactic acid to delay the emergence of fatigue.
