解脲支原体液体培养及电镜观察 被引量:2

摘要 目的:用液体培养基分离解脲支原体(UU),并电镜下观察其形态特征。方法:用选择性UU液体培养基分离尿生殖道标本中的UU,阳性结果经PCR方法证实,电镜下观察其形态。结果:阳性者为尿素酶阳性,培养液呈透明红色,PCR检测为阳性,电镜观察可见大小不等、无细胞壁的圆形、椭圆形、杆状、发芽状多形性特征的UU细胞,大小为0.03~0.12μm×0.12~0.35μm,未见细菌细胞。结论:选择性液体培养基能充分利用UU在液体培养基中生长迅速、分解尿素和缺乏细胞壁的特性,有效地抑制尿生殖道标本中的细菌生长,获得UU纯培养物。液体培养法操作简单,重复性好。 Objective:To isolate Urcaplasma urcalyticum(UU) and observe their cell strustures under electron microscopy(EM).Method:U.urealyticum was isolated from urogenital tract specimans with a selective broth medium.Positive cultures were confirmed with PCR test for U.urealyticum and observed by electron microscipe.Result Positive cultures showed urease positive and red transparent changes and positlve result of PCR for UU.Electronmicroscopically,UU realyticum showed polymorphic character of spherical,oviform,rod,budded shape and 0.03~0.12μm by 0. 12~0.35μm in size and lack of cell wall.No bacteria was observed.Conclusion:Selective broth medium can take advantage of UU realyticum's fast growth in the broth medium,urease positive,and lack of a true cell wall effectively suppressed bacteria growth in the specimens and may get the pure cultures of U urealyticum.The cultur method of selective broth medium is easy to operate with good repetition.
