同种异体骨髓间充质干细胞复合胶原海绵修复兔桡骨缺损的实验研究 被引量:2

摘要 目的:了解兔骨髓间充质干细胞(BMscs)同种异体移植的存活分布情况及同种异体兔BMscs复合胶原海绵(CS)情况,观察其对兔桡骨1.5 cm缺损的修复效果,为将来的进一步应用奠定实验基础。方法:采集、培养、纯化扩增新西兰大白兔BMscs,用eGFP荧光标记BMscs,与胶原海绵联合培养,植于同种异体动物臀大肌,观察存活及分布情况。建立兔桡骨缺损(1.5 cm)模型,45只新西兰大白兔随机分为3组:同种异体BMscs/CS组(A组),单纯胶原海绵组(B组),空白对照组(C组)。术后分阶段行X线、组织学检查及生物力学检查,评价其修复效果。结果:eGFP标记的BMscs异体移植存活时间超过3周。X线示12周A组大白兔5只中有4只达到骨性连接。新生骨量呈A组-C组递减,至12周无一愈合。随着时间推移,A组编织骨髓腔和骨小梁增多,修复过程优于B、C组。A组生物力学指标均低于健侧正常对照组,但各指标能达到正常组的80%~90%。结论:同种异体MSCs复合CS可以修复1.5 cm兔桡骨缺损。 Objective:To study the survival and distribution of allogenic mesenchymal stem cells(BMscs) after implantation into other rabbits,evaluate osteogenetic effectiveness of collagen-sponge mixed with allogenic BMscs of rabbit in repairing of radial shaft defects,and lay the foundation for further application.Methods: The BMscs of New Zealand white rabbits were collected,cultured,purified and amplified,they were marked with eGFP,cultured with collagen-sponge and planted into ectogluteus of other rabbits in order to observe the survival and distribution of BMscs.Model of radial bone defects(15 mm)of rabbits was established.45 rabbits were divided into 3 groups,group A(BMscs/CS group),group B(CS group) and group C(untreated group).Roentgenographic,histologic,biomechanics indexed of the animals were examined at certain time after surgery to evaluate osteogenetic effectiveness.Results: The BMscs marked with eGFP could survive for 3 weeks.On the 12th week,4 of 5 radial bone defects healed in group A.The quantity of callus decreased from group A to C.In group B and C,no one rabbits healed on the 12th week.Biomechanics result showed the indexes of group A were lower than the opposite side normal control groups.Conclusion: BMscs mixed with CS can repair radial defect shorter than 1.5 cm in rabbit.
