摘要
目的优化分光光度法测定橙皮苷含量的实验方法和桔实中双氢黄酮类橙皮苷的提取工艺。方法分析方法:采用L9(34)正交实验,考察溶剂类型、显色时间、温度和显色剂用量等影响因素;提取工艺:采用L9(34)正交实验,考察乙醇浓度、溶剂用量、提取时间和提取温度等影响因素;参照Fenton反应原理研究不同桔实中黄酮的清除.OH中作用。结果分光光度法测定橙皮苷含量的最佳实验条件:橙皮苷与5%A l(NO3)3溶液(m:V),在20℃反应20 m in,最大吸收波长为400 nm;橙皮苷提取的最佳工艺条件:70%乙醇为提取溶剂,采用料液比1∶20(m∶V),在50℃下中速搅拌提取3 h。桔实中黄酮提取液对Fenton体系产生的.OH有一定的清除能力。结论优选得到的提取工艺提取不同桔实中的橙皮苷,发现橘皮中橙皮苷含量最高。
Objective To study the determination method of hesperidin content by spectrophotometer and extraction technology of hesperidin from peels of citrus.Methods Analysis technology: The type of solvent,temperature,quantity of indicator and the time of reaction were investigated through orthogonal tests of L9(34);Extraction technology: The concentration of ethanol,quantity of ethanol,time and temperature of extraction were investigated through orthogonal tests of L9(34).In addition,the scavenging effect of flavanone from peels of citrus on hydroxyl radical was studied.Results The optimum technical conditions for determining the content of hesperidin by spectrophotometer were that: hesperidin was immersed with 5% Al(NO3)3(m:V)at 20℃ for 20 min and the maximum absorbance wavelength was 400 nm;the optimum technical conditions of extracting hesperidin were that: 70% ethanol was used as solvent,and the hesperidin was extracted at 50℃ for 3 h when the powers was stirred at medium-speed,the mass ratio of peels weight to 70% ethanol solution was 1:20(m:V).In addition,total flavanone from peels of citrus had effects on scavenging of hydroxyl radicals.Conclusion The content of hesperidin in peels is the highest by improved extraction technology.
