摘要
目的研究芎汤不同提取部位对小鼠急性心肌缺血的保护作用。方法分别制备芎汤乙醇提取液过大孔吸附树脂柱的20%,40%,60%,95%乙醇洗脱部位(EE20,EE40,EE60,EE95部位)。用腹腔注射异丙肾上腺素造成小鼠急性心肌缺血模型,观察不同洗脱部位对心电图、红细胞膜流动性的影响。结果EE20,EE40,EE60部位对异丙肾上腺素引起的心肌损伤小鼠,可拮抗S-T段异常变化;EE20,EE40部位可显著性降低荧光探剂DPH标记的小鼠红细胞膜荧光偏振度,增强红细胞膜流动性。结论芎汤EE20和EE40部位对实验性心肌缺血有明显的保护作用。
Objective To explore the protective effect on acute experimental ischemic myocardium in mice by different elutes from macroporous resin for separation of Xiongqiong decoction.Methods The ethanol extract from Xiongqiong Decoction was obtained firstly.Then 20,40,60 and 95 percent ethanol elute(EE20,EE40,EE60 and EE95) were prepared by using the column stuffed with absorbent macroporous resin.Acute myocardial ischemic model induced by intraperitoneal administration of isoproteronol(ISO) in mice was adopted to observe the effects of the different elutes on the electrocardiogram and the membrane fluidity of erythrocytes.Results Compared with the model group,EE40 and EE60 could reduce the elevation of ST segment induced by ISO.EE20 and EE40 could decrease the fluorescence polarization which demonstrated the effect of increasing the cell membrane fluidity in the inactive state.Conclusion The extracts of EE20 and EE40 have protective effect against ISO-induced acute myocardial ischemia in mice.
