摘要
目的研究MS培养基中大量元素、微量元素对彭泽贝母愈伤组织生长及生物碱含量的影响,筛选有利于彭泽贝母愈伤组织生长和生物碱积累的培养基配方。方法采用正交和单因素实验设计,以生长40 d的彭泽贝母愈伤组织的生长率和生物碱含量为指标,评价各因素对其影响的大小。结果各因素对彭泽贝母愈伤组织生长影响大小依次为P>Ca>K>N,对生物碱积累影响大小依次为K>N>Ca>P;培养基中NO3-N含量较高时,有利于生物碱的积累;微量元素缺乏时均有利于彭泽贝母愈伤组织的生长,缺Cu、缺I时不利于生物碱的积累。结论有利于彭泽贝母愈伤组织生长和生物碱积累的最佳组合分别为:N3P1K3Ca2、N1P2K3Ca1;培养基中大量元素、微量元素对彭泽贝母愈伤组织生长及生物碱含量存在一定影响。
Objective To study the effect of large elements,trace elements of MS culture medium on the growth and alkaloid content of Fritillaria monantha Migo callus,and to select the culture medium which is advantageous in its growth and alkaloid accumulation.Methods With the growth rate and alkaloid content of the 40 days' callus as indexes,single-factor and orthogonal experimental design was conducted to evaluate the impact of various factors.Results The effect of various factors on the growth of the callus is P〉Ca〉K〉N,and on the alkaloids was K〉N〉Ca〉P;the alkaloid content was higher when the proportion of NO3-N was higher than NH+4-N;Lacing trace elements were all advantageous in growth of the callus,but lacing I and Cu were not helpful to the accumulation of alkaloids.Conclusion The best combination of the various factors for the growth and the accumulation of alkaloids of the callus respectively is: N3P1K3Ca2、 N1 P2 K3Ca1;large elements,trace elements of MS culture medium have effects on the growth and alkaloid content of Fritillaria monantha Migo callus.
