多西紫杉醇对人舌鳞癌Tca8113细胞的生长抑制作用

摘要 目的研究多西紫杉醇对人舌鳞癌Tca8113细胞增殖及凋亡的作用。方法将0.1μmol/L的多西紫杉醇分别作用于Tca8113细胞12、24、48h,分别用倒置显微镜观察细胞形态、流式细胞术检测细胞凋亡状况;将浓度为0.001、0.01、0.1、1、10μmol/L的多西紫杉醇分别作用于Tca8113细胞12、24、48h后,用MTT法检测细胞增殖状况。结果多西紫杉醇对Tca8113细胞的增殖有明显的抑制作用,且浓度在0.001、0.01、0.1、1、10μmol/L时呈时间和浓度依赖性;镜下可见典型的凋亡细胞形态学特征。流式细胞术检测:多西紫杉醇浓度为0.1μmol/L时,作用12、24、48h,凋亡率增加,细胞阻滞于G2/M期。结论多西紫杉醇对Tca8113细胞有增殖抑制和诱导凋亡作用,这可能与抗癌机制有关。 Objective To explore the effects of docetaxel on the growth and apoptosis of tongue cancer cell line Tca8113. Methods Tongue cancer cell line Tca8113 was cultured with 0.1 μmol/L docetaxel for 12,24 and 48h,respeetively. The cell morphology was observed under an inverted microscope and the cell apoptosis was detected by flow cytometry. Tongue cancer cell line Tca8113 was cultured with docetaxel in 0.001,0.01,0.1,1 and 10μmol/L for 12,24 and 48h,respectively. The cell growth was detected by MTT. Results The cell proliferation showed a dose-and time-dependent decrease after cultured with docetaxel in 0.001,0.01,0.1,1 and 10μmol/L.A typical feature of apoptosis was observed under microscope. The flow cytometry showed that the apoptosis rate increased after adding 0.1 μmol/L docetaxel for 12,24 and 48h respectively. Tongue cancer cells were arrested in G2/M. Conclusion Docetaxel can inhibit the growth of tongue cancer cell line Tca8113 and induce its apoptosis,whieh might be one of anti-cancer mechanisms.
