转染p21^(WAF1/CIP1)对肾癌细胞系GRC-1促凋亡作用

Apoptosis induced by p21^(WAF1/CIP1) gene transfection on renal carcinoma cells

目的：研究转染p(21WAF1／CIP1)对肾癌的促凋亡作用．方法：采用脂质体介导的逆转录病毒载体将p21(WAF1／CIP1)转入无P21蛋白表达的肾癌细胞系GRC－1中，通过电镜、流氏细胞仪、DNA梯电泳检测细胞凋亡．结果：电镜检测到凋亡细胞，DNA电泳呈梯状．所检测细胞中15．3％出现凋亡．结论：p21(WAF1／CIP1)过表达有促进GRC－1细胞凋亡作用．

Aim: To study the role of p21WAF1/CIP1 genetransfection on renal carcinoma cells apoptosis. Methods:transelectronmicroscope. flowcytometry and DNA elctophoresiswere used to observe cells apoptosis which induced bylipofection-mediated p21WAF1/CIP1 transfection in GRC- lcells.Results: Apoptosis cells could be seen by electron microscope,the 'DNA ladder' appeared on DNA electrophoresis, and anapoptotic peak appeared in front of G1 phase,the apoptoticcells accounted for 15. 3%. Conclusion: It confirmed thatthe overexpression of p21WAF1/CIP1 can induce apoptosis inGRC-1 cells.
