摘要
eIF4E(eukaryotic translation initiation factor4E)家族基因在多种病毒侵染植物过程中起重要作用。应用RT-PCR和RACE方法在番木瓜中克隆了cDNA全长为995bp的eIF4E异构体基因,命名为CP-eIF(iso)4E(GenBank登录号为FJ595992)。其编码的蛋白含有206个氨基酸,蛋白一级结构中的活性部位位点高度保守,三维结构中可能的抗性位点位于帽结合区域(cap-binding pocket)表面。半定量RT-PCR结果表明,CP-eIF(iso)4E在番木瓜叶片中表达较弱,在花瓣中表达最强。
eIF4E (eukaryotic translation ruination tactor 4E) famliy genes pray important roles against virus from infecting plants. A complete cDNA of 995 bp from papaya was cloned using the method of RT-PCR and RACE. It was a isoform eIF4E, so it was named CP-elF(iso)4E (GenBank accession No. FJ595992). The predicted CP-eIF (iso) 4E protein had 206 amino acids, in which the active site was high conservative in primary structure, and the predicted virus-resistance site was located on the surface of cap-binding pocket in 3-D structure. Semi-quantitative RT-PCR analysis indicated that CP-eIF (iso) 4E expression was lower in leaf and higher in petal of papaya.
