NRSE/RE-1序列与小鼠胚胎干细胞向神经细胞诱导分化的关系

摘要 目的探索神经限制性沉默元件(NRSE/RE-1)序列与小鼠胚胎干细胞向神经细胞诱导分化的关系。方法应用序贯诱导法定向诱导鼠胚胎干细胞分化为神经细胞,基因芯片筛选出的11种含NRSE/RE-1序列的神经相关基因,实时定量PCR测定其在分化过程中的表达。结果诱导分化后的胚胎干细胞表达多种成熟神经元标志,11种含NRSE/RE-1序列基因随诱导分化的进程表达量升高。结论鼠胚胎干细胞可通过序贯诱导分化为神经细胞,NRSE/RE-1序列可能是胚胎干细胞向神经元诱导分化的主要调控位点。 Objective To explore the relationship between NRSE/RE-1 and induced differentiation from mouse embryonic stem ceils (ESCs) to neural cells. Methods Sequential induction was used to induce the differentiation of mouse ESCs to neural cells. Eleven kinds of neural genes with NRSE/RE-1 sequence screened from microarray during differentiation were detected by real-time quantitative PCR. Results The induced ESCs began to express some markers of mature neurons,and the expression of 11 kinds of genes with NRSE/RE-1 sequence increased during differentiation. Conclusion The mouse ESCs can be induced to neural cells by sequential neural induction, and the NRSE/RE-1 sequence may be the primary regulation site during the indued differentiation.
