摘要 目的探讨胰腺导管腺癌中Smad特异性E3泛素蛋白连接酶1(SMURF1)基因表达水平的异常情况。方法以相应癌旁正常组织为对照,应用半定量RT-PCR方法检测31例胰腺导管腺癌组织中SMURF1 mRNA表达水平;应用Western blot方法检测相应标本中SMURF1蛋白水平的异常情况。结果在31例病例中,肿瘤组织SMURF1 mRNA表达高于相应癌旁正常组织的有20例,统计分析结果肿瘤组与对照组差异显著(P<0.01);其中19例中同时存在SMURF1蛋白的高表达(P<0.01)。结论胰腺导管腺癌中SMURF1 mRNA和蛋白表达水平升高,这可能是胰腺癌发生发展的另一新的分子机制。 Objective To assess the abnormal expression level of Smad-ubiquitination regulatory factor 1 (SMURF1) gene in pancreatic ductal adenocarcinoma. Methods With the paired normal tissue as a control, semi-quantitative RT-PCR was used to detect the expression level of SMURF1 mRNA in 31 cases with pancreatic ductal adenocarcinoma. The SMURF1 protein expression level in these tissues was determined by Western blot. Results In 31 cases,20 cases showed higher expression level of SMURF1 mRNA and 19 cases showed higher expression levels of SMURF1 protein in tumor group compared with that in control group and the differences between two groups was statistically significant (P 〈 0.01 ) . Conclusion The SMURF1 mRNA and protein expressed highly in pancreatic ductal adenocarcinoma. This may be a new molecular mechanism in the development and progression of pancreatic carcinoma.
