围产期低剂量三丁基锡暴露对子代雌性小鼠发育及雌激素水平的影响 被引量:1

Effects of Perinatal Exposure to Low-dose Tributyltin on Development and Estrogen Level of Female Offspring Mice

摘要 目的探讨围产期低剂量三丁基锡(TBT)暴露对雌性昆明小鼠生长发育及血清雌激素的影响。方法将清洁级健康成年妊娠昆明小鼠随机分为100、10、1μg/kgTBT染毒组和对照组(玉米油),每组6只。从妊娠第6天起,采用经口灌胃方式进行染毒,每天1次,染毒剂量为1ml/kg,直至哺乳期结束。出生后第49天,每组随机抽取10只雌性仔鼠,称重后取血,分离子宫及卵巢,称重,计算子宫及卵巢的脏器系数。采用放射免疫法测定血清中雌二醇的浓度。结果与对照组比较,各TBT染毒组雌性仔鼠睁眼时间延长,阴道张开时间提前,差异均有统计学意义(P<0.05或P<0.01);且随着TBT染毒剂量的增加,雌性仔鼠睁眼时间呈延长趋势。各TBT染毒剂量组雌性仔鼠体重、子宫重量及其脏器系数间比较,差异均无统计学意义(P>0.05)。与对照组比较,10μg/kgTBT染毒组雌性仔鼠血清中雌二醇的浓度和卵巢重量及其脏器系数升高,差异均有统计学意义(P<0.01);且仔鼠血清雌二醇水平随着染毒剂量的增加呈递增趋势。结论围产期低剂量TBT暴露对雌性仔鼠有内分泌干扰作用,影响其生长发育,导致血清雌激素水平升高。 Objective To explore the effects of perinatal exposure to low dose tributyhin (TBT) on development and estrogen level of female offspring KM mice. Methods The CL healthy adult pregnant KM mice were randomly divided into 4 groups, 6 in each group,they were given doses of TBT (100,10 and 1 μg/kg ) by gavage from days 6 of gestation to the end of lactation,1 time each day, at a volume of 1 ml/kg. On postnatal day (PND) 49, 10 female offspring mice were randomly selected in each group and killed after weighed and the blood was collected. The uterus and ovary were weighed for account of viscera coefficient. The concentrations of estrogen in serum were measured with radioimmunoassay. Results Compared with the control group the time of eye opening was significantly delayed and the vagina opening was significantly ahead in treatment groups (P〈0.05, P〈0. 01 ), and there was an increasing-trend in time of eye opening following the TBT dose increasing. There were no significant differences in the body weights of PND21, PND49 and the gains of body weight, and the weights of uterus and ovary and their coefficients (P〉0.05). The weight of ovary and its coefficient increased significantly in 10 μg/kg group (P〈0.01). There was an increasing-trend in serum estrogen following the TBT dose rising, and a significant increase in 100 μg/kg group (P〈0.01). Conclusion Perinatal exposure to low-dose TBT may have endocrine disrupting effect on the female offspring mice, and affect their growth and development.
