人垂体肿瘤转化基因在子宫内膜癌发生发展中作用的研究 被引量:5

摘要 目的探讨人垂体肿瘤转化基因(hPTTG)蛋白表达和上皮细胞钙黏蛋白(E-cad)异常表达在子宫内膜样腺癌发生、发展过程中的意义。方法采用免疫组化法PV-9000检测46例子宫内膜样腺癌、8例子宫内膜不典型增生、11例子宫内膜单纯性增生以及17例正常子宫内膜组织中hPTTG及E-cad蛋白的表达水平,并将结果进行分析。结果hPTTG蛋白在子宫内膜样腺癌组织中呈过度表达,且hPTTG蛋白阳性表达率随着子宫内膜样腺癌的手术-病理分期期别增高、组织学分级降低、肌层浸润及淋巴结转移的发生而升高(均P<0.05)。E-cad蛋白在17例正常子宫内膜组织中均为正常表达,在子宫内膜不典型增生及子宫内膜样腺癌组织中,其异常表达率明显升高,且E-cad蛋白的异常表达率随着子宫内膜样腺癌的手术-病理分期期别增高、组织学分级降低、肌层浸润及淋巴结转移的发生而升高(P<0.05)。hPTTG蛋白阳性、阴性的子宫内膜样腺癌组织中E-cad蛋白的异常表达率分别为71.0%,20.0%,差异有统计学意义(P<0.01),子宫内膜样腺癌组织中E-cad蛋白的异常表达和hPTTG蛋白的表达呈正相关(r=0.486)。结论hPTTG蛋白表达和E-cad蛋白异常表达均促进子宫内膜样腺癌的发生和浸润演进过程,对子宫内膜癌的诊断和预后判断有重要意义。 Objective To investigate the role of aberrant pituitary tumor-transforming gene(hPTTG)and epithelia-cadherin(E-cad) expression in the tumorigenesis and progression of endometrial adenocarcinoma.Methods Immunohistochemistry was employed to detect the expression of hPTTG and E-cad protein in 46 cases of endometrial adenocarcinoma,8 cases of endometrial dysplasia,11 cases of simple hyperplasia,17 cases of normal endometrium tissues.Comparison of their expression was performed with clinicopathological parameters of carcinomas. Results The hPITG protein was overexpressed in 67.4% of endometrial adenocareinomas, mole frequently than in simple hyperpla- sia of endometrium ( 18.2%, P 〈 0.05 ). It was related to the surgical-pathological stage, the grade of cellular differentiation, the depth of myometrial invasion, and lymph node metastasis of adenoeareinoma (P 〈 0.05 ).The normal expression of E-cad protein was found in all of 17 eases of normal endometrial tissues. The abnormal expression rates of E-cad protein in the endometrial dysplasia and adenoeareinoma were higher than those in normal endometrium and simple hyperplasia ( P 〈 0.05 ). It was correlated with file surgical-pathological stage, the grade of cellular differentiation, the depth of myometrial invasion and lymph node metastasis of these adenoeareinomas. The abnormal expression rate of E-cad protein was significantly higher in bPPPG-positive than -negative endometrial adenocarcinomas (71.0% vs 20.0%, P 〈 0.01 ). The abnormal expression of E-cad protein was closely correlated with the expression of hPTYG protein in endometrial adenoeareinoma( r =0.486 ). Conclusion The expression of hPTTG protein and abnomlal expression of E-cad protein was involved in the carcinogenesis and progression of endometrial ad They were of great value to evaluate the outcome of endometrial adenocarcinoma patients .
