摘要 目的:探讨羟基磷灰石纳米粒子(Nano-HAP)对人胃癌细胞系增殖、侵袭的影响及机制。方法:以0、25、50、100、150、200μg/mL的Nano-HAP分别作用3种细胞48h;100μg/mL的Nano-HAP分别作用3种细胞细胞12、24、36、48、60、72h,使用MTT法检测Nano-HAP对胃癌细胞增殖的影响;100μg/mL Nano-HAP分别作用于SGC-7901、BGC-823、MKN-28细胞48h,以倒置相差显微镜观察细胞的生长情况;以过河实验、黏附测定观察Nano-HAP对胃癌细胞侵袭力的影响。结果:Nano-HAP可明显抑制胃癌细胞的增殖和生长,且呈剂量-时间效应关系。过河实验、黏附实验显示,Nano-HAP能降低胃癌细胞的体外侵袭和运动能力,随浓度增高,过河时间延长,细胞黏附力降低(P<0.05)。结论:Nano-HAP可抑制人胃癌细胞的增殖和生长,并降低其体外侵袭能力。 Objective To study the effect and mechanism of action of Nanoscale hydroxyapatite on proliferation and invasion of gastric cancer cells. Methods SGC-7901,BGC-823 and MKN-28 cells were incubated with 0,25,50, 100,150, and 200 μg/mL Nano-HAP for 48 h or incubated with 100 μg/mL Nano-HAP for 12,24,36,48,60,and 72 h. Tetrazolium-based eolorimetrie assay was used to evaluate cell growth. Gastric cancer ceils were incubated with 100 μg/mL Nano-HAP for 48 h phase contrast light microscopy was used to evaluate cell growth. The crossing-river test, adhesion experiment were used to examine the invasivedness of gastric cancer cells. Results Nano-HAP inhibited the growth and proliferation of gastric cells in a dose-and time-dependent manner, adhesion experiment showed that Nano-HAP decreased the in vitro invasion of gastric cancer cells and their crossing-river time was increased significantly. Conclusions Nano- HAP inhibits the in vitro proliferation and growth of human gastric cancer cells.The anti-invasive activity may be the results of decrease cell motility.
