血红素氧合酶-1在远端缺血预适应的大鼠心肌中的作用探讨

摘要 目的:通过对下肢缺血预适应后急性心肌梗死大鼠心肌梗死面积和炎症反应变化的观察,探究血红素氧合酶-1(HO-1)在心肌保护中的作用机制。方法:将64只健康雄性成年Spraque-Dawley(SD)大鼠随机分为急性心肌梗死组和下肢缺血预适应组。各组大鼠分别在急性心肌梗死后2、4、6、12h四个时间点处死取材。经冷冻、切片后用2%TTC法染色,于手术显微镜下仔细分离梗死部位(IS)和缺血危险区(AAR),并计算IS与AAR重量比值(IS/AAR),推测心肌梗死面积。双抗体夹心(ELISA)法测定血清白细胞介素6的含量,C反应蛋白浓度测量使用速率散射比浊法。用全自动生化分析仪检测肌酸激酶(CK-MB)含量。采用逆转录-多聚酶链反应(RT-PCR)测定心肌组织中HO-1 mRNA表达情况。结果:下肢缺血预适应组HO-1 mRNA表达升高,心肌梗死面积比急性心肌梗死组明显减小;CK-MB含量明显下降。结论:HO-1对远端器官缺血预适应后的心肌有保护作用。 Objective To observe expression of berne oxygenase-i (HO-1) mRNA and changes of inflammatory reaction in Sprague-Dawley rats with acute myocardial ischemia (AMI) after ischemic preconditioning for limbs. And to investigate regularity of HO-1 mRNA expression and its dynamic changes in early ischemic myocardium, also the protective effects on ischemic myocardium. Methods Male Sprague-Dawley rats were randomly divided into two groups: AMI group and limb isehemic preconditioning group(LIP). All the rats both in the AMI and LIP group were sacrificed at 2, 4, 6, 12 hours after AMI. We recorded the 2-lead electrocardiogram of all the rats. Myocardial infarct sizes were examined by Evans and triphenyl tetrazolium chloride. Levels of C reactive protein in serum were measured by rate nephelometry and ereatine kinase-MB (CK-MB) in serum was measured by automatic biochemistry analyzer. We determined the serum levels of interleukin-6 using enzyme linked immunosorbent assay and the expression levels of HO-1 mRNA in myocardium were evaluated by reverse transcription-polymerase chain reaction. Results The expression of HO-lmRNA was increased up in LIP group, the myocardial infarct size was decreased significantly in LIP group that compared with AMI group,the content of CK-Mb was also obviously decreased. Conclusions HO-1 can protect the infarcted myocardial after isehemic preconditioning for limbs in rats.
