骨髓基质细胞对微血管内皮细胞增殖、迁移、管腔化的影响

摘要 目的:观察骨髓基质细胞(BMSC)对微血管内皮细胞的增殖、迁移、血管新生的影响并探讨其作用机制。方法:体外培养BMSCs,通过体外BMSC/微血管内皮细胞共同培养模型,在倒置相差显微镜下,利用图像分析系统观察BMSCs条件培养基对微血管内皮增殖、迁移的影响,利用细胞外基质管腔形成实验评价BMSCs对微血管内皮细胞血管新生的影响。结果:BMSCs上清条件培养基、BMSCs/微血管内皮细胞共培养上清条件培养基与对照组比较可促进微血管内皮细胞增殖[(0.165±0.011)vs(0.170±0.008)vs(0.152±0.010),P<0.05]、划痕修复能力[(1850.269±32.64)μmvs(2025.420±134.300)μmvs(1695.891±32.073)μm,P<0.05]和管腔形成能力[(3.69±0.28)mm/mm2vs(6.59±0.24)mm/mm2vs(1.47±0.21)mm/mm2]。结论:BMSCs分泌的因子可以促进微血管内皮细胞增殖、细胞迁移和血管新生。BMSCs移植治疗脑缺血,不仅通过神经保护起作用,也通过影响微血管内皮细胞,促进血管新生发挥治疗作用,改善神经功能。
