白筘叶总黄酮的大孔树脂纯化工艺研究 被引量:5

摘要 目的:研究白簕叶总黄酮的大孔树脂纯化工艺。方法:通过静态、动态吸附从8种大孔树脂中确定白簕叶总黄酮精制最优树脂为HPD-600,以黄酮吸附率、洗脱率、黄酮纯度和薄层色谱结果为评价指标,综合评判纯化最优工艺。结果:其最佳吸附工艺参数为:上样量30mL,上样浓度4mg·mL-1、上样液pH5~6、吸附流速0.5mL·min-1;最佳洗脱条件为:洗脱液体积40mL,pH5~6,70%的乙醇溶液,洗脱流速2mL·min-1,精制后黄酮质量分数超过50%。TLC结果显示,不同浓度的洗脱液中均含有芦丁和金丝桃苷,pH1~2、3~4、5~6的洗脱液均含有芦丁和金丝桃苷。结论:采用HPD-600树脂精制白簕叶总黄酮,操作简单,成本低廉,精制效果突出,有较高的工业生产应用价值。 A study on the adsorption of total fiavonoids from the leaves of A. trifoliatus leaves has been conducted and HPD-600 out of 8 kinds of different macro porous resin is the optimal material for the adsorption.Judging by adsorption rate of flavonoids, desorption rate, purity and TLC results,the optimal adsorption process parameters were as follow-sample volume of 30mL, sample concentration of 4mg · mL^-1, sample pH5-6, and adsorption velocity of 0.5mL · min^-1.The best desorption conditions were 40mL elution solution,pHSN6,70% ethanol solution, adsorption velocity of 2mL · min^-1.This method can achieve purity more than 50%. Rutin and hyperoside were detected by TLC method both in elution solution of different concentrations or pH values(1-2,3-4,5-6).The study can finally concluded that the purification method of total flavonoids in A.trifoliatus leaves with HPD-600 resin had the advantage of simple operation process,low cost and high level of purification,it could have a great potential in industrial production of A.trifoliatus flavonoids.
