摘要
目的:以人白血病细胞系HL60和小鼠B16BL6黑色素瘤细胞株为对象,以细胞抑制率、细胞凋亡为指标,评价白花蛇舌草提取物及多糖的体外诱导癌细胞凋亡效应,为白花蛇舌草的抗肿瘤效应提供实验依据。方法:采用水提取物、乙醇提取物及多糖提取物,应用MTT和XTT法检测提取物对B16BL6、HL60肿瘤细胞株的细胞抑制率,采用FCM法检测其诱导细胞凋亡的效果。结果:(1)MIT/XTT法定量检测,3种提取物对肿瘤均有一定的抑制作用,且抑制作用随药物浓度的增加而增大;(2)FCM法检测,白花蛇舌草醇提物和多糖对B16BL6细胞凋亡和细胞周期的影响与空白组比较差异有显著性。结论:白花蛇舌草水提物、醇提物和多糖对小鼠B16BL6黑色素瘤细胞和HL60细胞株均具有明显的抑制作用,醇提物和多糖具有明显的诱导细胞凋亡作用。
OBJECTIVE Take the HL60(Human promyelocytic leukemia cell line) and B16BL6(mouse melanoma cell line) as the object,and the inhibition ratio and the apoptosis of the cells as the indicatrix, the purpose was estimate the effect of the extraction and the polysaccharide of Oldenlandia diffusa. METHODS Aqueous extract, alcohol extract and polysaccharide were applied to the B16BL6 and HL60. Then the cells inhibition ratio was detected by MTT method and the apoptosis of ceils by FCM (flow cytometry) method. RESULTS (1) In MTT quantitate detection, the extractives of Oldenlandia diffusa had the inhibitive function to tumors propertional to the concentration. (2) The alcohol extract and polysaccharide of Oldenlandia diffusa have the effect of the apoptosis and the cell cycle of B16BL6 by FCM method. CONCLUSION The aqueous extract, alcohol extract and polysaccharide of Oldenlandia diffusa have the remarkable inhibitive function to the HL60 and B16BL6 . The alcohol extract and polysaccharide have the remarkable function to induced the apoptosis of the cells.
